Research Database

From the Peptide Explorer

A potent, selective, cell-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT is overexpressed in obese adipose tissue and ageing muscle, where it depletes the methyl donor SAM and generates the metabolically inert 1-methylnicotinamide. Inhibiting NNMT restores SAM and NAD+ precursor pools, reactivating lipolysis and myogenesis.

Key Mechanisms

  • Selective NNMT enzyme inhibition
  • Restoration of SAM (methyl donor) levels
  • Increased NAD+ precursor availability
  • Enhanced adipocyte lipolysis
  • Stimulation of satellite-cell-driven myogenesis

Primary Research Areas

  • Diet-induced obesity models
  • Sarcopenia and muscle regeneration
  • NAD+ metabolism
  • Methyl-donor biochemistry

Key Research Findings

  • Reduced white adipose mass and improved insulin sensitivity in diet-induced obese mice (Neelakantan et al., 2018)
