From the Peptide Explorer
A potent, selective, cell-permeable small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT is overexpressed in obese adipose tissue and ageing muscle, where it depletes the methyl donor SAM and generates the metabolically inert 1-methylnicotinamide. Inhibiting NNMT restores SAM and NAD+ precursor pools, reactivating lipolysis and myogenesis.
Key Mechanisms
- ›Selective NNMT enzyme inhibition
- ›Restoration of SAM (methyl donor) levels
- ›Increased NAD+ precursor availability
- ›Enhanced adipocyte lipolysis
- ›Stimulation of satellite-cell-driven myogenesis
Primary Research Areas
- ›Diet-induced obesity models
- ›Sarcopenia and muscle regeneration
- ›NAD+ metabolism
- ›Methyl-donor biochemistry
Key Research Findings
- Reduced white adipose mass and improved insulin sensitivity in diet-induced obese mice (Neelakantan et al., 2018)
- Restored myogenic capacity and muscle mass in aged mouse models (Dimet-Wiley et al., 2022)
- Depletion of 1-methylnicotinamide and restoration of intracellular SAM
